What starch damage content can be measured using the Starch Damage Assay Kit (K-SDAM)? What is the linear range of the glucose measurement?

The glucose measurement (incubation with GOPOD Reagent) is linear between 4 and 150 micrograms of glucose per assay, which is equivalent of 0.3 to 12% of damage starch in samples, when the method is followed as described in the data booklet.

 

The absorbance received for your sample must not exceed the absorbance values obtained for 150 micrograms of glucose standard (0.1 mL of standard at 1.5 mg/mL that is supplied with the kit).

 

If an expected starch damage content in your sample is higher than 12% or received absorbance values for the samples are higher than the absorbance values obtained for the glucose standard, the sample solution must be diluted sufficiently with 0.2% sulphuric acid after centrifugation at step 6, and repeated from step 7 of the assay procedure.


Samples between 12 - 50% should be diluted 1:5 and sample between 50 - 100% should be diluted 1:11 before  amyloglucosidase treatment. The dilution should be accounted for in the calculations.
